KHV (Koi Herpes Virus) Diagnostics

The infection shows up at various times during infection. The virus might be noticeable in the gills or blood for just a couple of hrs or days. In the liver and especially the kidney, contaminated fish will typically have detectable infection.

So, not all cells kinds are “equal” diagnostically.

Best: Produce and homogenize kidney and liver. Ship frozen or near freezing for PCR. Formalin repair for Sitting Hybridization.

Handling Tissues

Long-dead (rotting) fish will certainly not work as the DNA stability can be jeopardized.

You should utilize real-time fish or EXTREMELY just recently dead fish for ideal results and use kidney, liver and gill.

PCR test tries to find particular DNA pens which are normally present in dead and perishing fish organs. Can be iced up.

Live infection will not be recoverable from icy samplings.

Gill “swabs” are just about pointless unless there is overt (apparent) disease amongst fish being tested.

Tests to Run

For now, the very best type to locate infection is a PCR examination.

PCR = Polymerase chain reaction examination. It identifies a precise sequence of unique nucleic acids only located in the Koi Herpes Virus.

Sitting Hybridization– Trying to find infection in the cells. (Showing infection).

There is also an agglutination (antibody) test for fish blood which would certainly show past infection, and assist display real-time fish.

Sitting Hybridization.

Operate on Tissues Seeking Carriers.

Detects infection DNA really IN the cells, which is a lot more specific and delicate compared to the PCR examination and the fish don’t have to be infected or passing away. Yet they probably need to die for example collection.

Tissues are prepped for histopathology, are stained with a noticeable healthy protein which expecteds specific viral KHV DNA, then the protein/DNA complex turns up blue under the microscopic lense.

This examination is offered at UGA and is good for virus spotting “carriers”.

Serological Assay.

This examination uses blood so the fish do not need to perish.

They’re presenting a marked KHV infection bit which triggers antibodies in the blood to agglutinate.

If the blood proteins agglutinate, it implies the fish has antibodies, and this suggests the fish was, in the last 8 months, affected.

Antibody “memory” to KHV appears only to last 6-8 months.

This is a good life saving test to say to if a fish was ever before affected with KHV.

Do the examinations “Play” together?

PCR searches for the infection LIVING in cells. Generally this virus test can not be relied upon to show provider states.

Sitting Hybridization searches for virus “HIDING” in the tissues and is far more delicate, equally as specific, and great for locating providers.

The Serology test confirms a fish made it through an infection. It doesn’t determine providers. It helps when used WITH sitting hybridization–.

To wit: If the serology shows ‘infection survivor’ then the in situ hybridization examination comes to be colossally essential to assist establish latency or service provider state.

In situ hybridization on a fish negative by serology is of far less worth. Why?

Due to the fact that if the fish is adverse on serology it would suggest it’s never been exposed or contaminated … So the second examination seeking virus PARTICLES is irrelevent.

Last year, Jeri Barnes’ Dover Township home was part of The Pond Experience — a self-guided tour of local ponds. This year, she said she wanted to visit other ponds.

Her first stop during the event Saturday was the courtyard next to York’s Central Market. She admired the new water feature — a small spring flowing between rocks, pebbles and plants.

Splash Supply Co. hosted the tour, which is in its second year. Sales associate James Pontz said about 300 people participated in 2013, and he expected a larger crowd Saturday.

Last year, he received lots of positive feedback, but some people couldn’t finish the tour. This year, he made it smaller, with nine stops centralized in York and along Route 83. Most offered samples from local businesses, live music and artists in action.

Jack Gettys and his wife, Linda, recently expanded their backyard oasis, tucked a few hundred yards from South Queen Street in York Township.

Gettys sold his Harley to finance recent projects, including a bog, waterfall, native plant area and bee hives. The couple’s koi pond now holds 4,500 gallons.

He started the pond eight years ago and has since joined the staff at Slash Supply Co.

On Saturday, several dozen folks filtered through the Gettys’ backyard area, which is shaded by a large Norway maple tree. Two local artists set up easels to paint the scene. An Aloha Snow truck served cool treats in the driveway.

Over in Spring Garden Township, York College business professors Chris and Mary Meisenhelter welcomed guests along the tour. The two began plotting their pond about a decade ago. It started as a flower bed and plastic tub. It now takes up most of their backyard. This year, they made the pond deeper and added another waterfall.

Mary noticed a survey on the National Wildlife Federation website and completed questions about the pond’s features. It’s now a Certified Wildlife Habitat.

Herons, deer, foxes and owls have visited the yard, Chris said. As of 1 p.m. Saturday, he said about 150 Pond Experience participants had stopped by.

Singer and guitarist Jef Woodall performed classic rock hits. Artists Robert Patierno and Janette Toth-Musser painted the pond scape. Both are members of the Pennsylvania Arts Experience, an organization that promotes visual art in the region. Proceeds from The Pond Experience will go to the PAE.

Toth-Musser spritzed her acrylic paint to keep it from hardening in the hot sun.

Patierno detailed his watercolors with ink, which dries quickly. He said he drew inspiration from the foot traffic, bubbling water and darting fish.

Pond tour visitors get inspired with watery gardens

By Heather Hacking

hhacking@chicoer.com @HeatherHacking on Twitter

CHICO >> Some of the most inspiring backyards for water-lovers had their gates wide open last weekend for the annual pond tour, a fundraiser for the programs of the New Vision Church of Chico.

This year marked the 20th anniversary of the event, which gives people a chance to borrow ideas, or even just to spend some tranquil moments enjoying the work of others.

Sandi Bosch and Maria Corporon have been attending the tour for about 10 years, and gain inspiration for their own yard.

At the home of Cathy and Craig Gurney, they loved the wildlife that joined visitors, as well as the aviary where the homeowners keep and rescue birds.

Something about ponds adds to the serenity of a place, Bosch noted.

From ashes to tranquil

The peace that comes from gurgling water is one of the reasons Rocky Campbell keeps a pond as the focal point of his backyard.

When he moved in, the land was raw. Over time he has built an intricate path to tranquility.

Thirty mature koi swim out of reach from the stretch of raccoons that might otherwise try to take a swipe at them.

Other predators include herons and king fishers. The key is to have a pond 34 inches deep, which will allow the fish an escape from the most common predators, Campbell explained.

Campbell also pointed out the water lilies, which begin to open when rays of sunlight reach them. They only last a short time, so he makes time to enjoy each one.

The water in the pond recirculates and goes through a filter. From this, he pulls out material rich in fish waste, which can be used to fertilize the yard.

Also recycled are some of the raw materials for yard structures. The stepping stones are leftover from construction projects and a satellite dish is used as the roof of a gazebo.

Watery play land

A sign along the driveway to Rick Coletti’s house reads “slow, adults at play.”

While certainly serene during the tour, the stylized work of Coletti is built for entertaining and beauty.

“I love the tropics and I love Bidwell Park,” Coletti said. His backyard combines elements of both.

The pool is the biggest body of water, with a pile of rocks to climb, and a slide for the descent.

After a trip down the slide, visitors can swim across the pool, climb over to the hot tub, then hop onto a partially-submerged bar stool. Here they’ll find a window to the kitchen, where the host can serve drinks from the bar.

Eye-dazzling features include large stepping stones (attached to pillars anchored under water). The appearance is that the stones are floating on the water’s surface.

At the front door of the home, Coletti has a rock pathway with more stepping stones surrounded by water.

Built with time in mind

The Gurney’s showcase home also showed the fun they had “decorating” outdoors.

The home was built a century ago, and many old-time elements remain.

A vintage gas pump becomes garden art and rusted garden elements hang along the side of a shed.

A new addition is a giant agricultural disc, bronzed with time. A path leads through the oval and if you make sound while standing inside, you’ll hear an echo.

See video here: http://www.tout.com/m/5y4g4o

Funds for local causes

The fundraiser helps the New Vision Congregational Church continue its community programs, which include a food pantry for the needy, providing meeting space to 12-step programs and the Shalom Free Clinic. Over the years money from the pond tour has gone to the Boys and Girls Club, the Butte College Horticulture Scholarship and Butte County Hepatitis C Task Force.

Aquarium creates 'special love nest' for shy clownfish

By | Good News –

At Sea Life London Aquarium, aquarists have created a “special love nest” for a pair of shy clownfish.

While there’s been a recent breeding boom at the aquarium, the clownfish have yet to jump on the baby-making bandwagon.

“It could be several reasons,” aquarist Pete Williams says of their apparent “stage fright.”

“It could be disturbances from the other fish in the display. Just be the general passing of the public as well might discourage the animals from breeding. There’s several different factors, so this is sort of our last go.”

To encourage the female to lay eggs, the tank has been prepared to more closely replicate their natural habitat, with small rock lining its floor. A red velvet curtain provides privacy from visitors — and sets the mood.

All that’s missing is Marvin Gaye and some champagne.

“We have an excellent track record with our breeding programs here at the Sea Life London Aquarium but this pair of beautiful Pink Skunk clownfish have been proving a bit shy,” explains Williams.

“So far they haven’t mated, so we have designed what we hope will be the perfect setting to bring them together. We’re giving them total privacy away from other marine life — and with a specially fitted curtain — away from human eyes too. Hopefully with this custom-made environment in place, we can look forward to welcoming some clownfish babies in the not too distant future.”

Here’s a fun fact, according to the Sea Life website:

“In a group of clownfish inhabiting an anemone at any given time, the largest clownfish is always a female. The smaller fish are male. When the female dies, the largest male will change sex to take over the female’s role!”

Theft, Brecksville Road: Two Parma men and a Brooklyn man were caught trying to steal a Koi fish at Tricker’s just after midnight July 14. The Parma men, 18 and 19, had bought a Koi from Tricker’s, but it died 29 days later. When they went back to the business to ask for a replacement fish, the management told them they had a strict 30-day return policy. The Parma men and their accomplice were seen by a neighbor scooping up a Koi with a net and putting it in a bag. They were charged with felony theft and criminal trespassing. The Koi was returned and is valued at $1,000-1,500.

Seal pups rescued in Canada

VANCOUVER, BRITISH COLUMBIA (CBC News/CNN) — Four newborn seal pups, some with their umbilical cords still attached, are being cared for by the Vancouver Aquarium after being rescued from B.C. shores on Tuesday.

B.C. resident Mark Levagood and his daughter saw one of the exhausted seal pups struggling to get onto a rock on Ogden Point in Victoria.

The male pup, which has been named Tantalum by volunteers, was airlifted to the Vancouver Aquarium Marine Mammal Rescue Centre, where it is now recovering.

Manager Lindsaye Akhurst said the seal pup is doing fairly well, but had been through a lot before being rescued.

It’s birthing season for seals in B.C.

And that means it’s a busy time of year for Vancouver’s aquarium.

During harbour seal birthing season, newborns often get separated from their mothers and need a new home for a little while.

Several lost pups have been discovered this week.

The aquarium has taken care of more than 40 of these temporary guests in the last month.

Nudists go shark-spotting at Newquay's Blue Reef Aquarium

Comments (0)

There are few things more frightening than a close encounter with a man-eating shark.

But a tank full of the toothy fish combined with a large group of visiting nudists could perhaps top that.

On Thursday 10 a group of naturists wisited Newquay’s Blue Reef Aquarium to enjoy close encounters with razor-toothed sharks.

The visit, which was organised as part of Nudefest 2014, was believed to be one of the biggest ever nude group visits to a UK aquarium.

Nudefest is an annual event organised by British Naturism which attracts hundreds of naturists from across the country to the South West.

During their visit to Blue Reef, members took a tour around the displays, taking part in a series of talks and feeding demonstrations by the aquarium’s aquarists.

British Naturism’s Andrew Welch said: “Nudefest is a well-established and hugely popular event in the naturist calendar and we are delighted to have returned to Cornwall this year.

“Guests were very excited by the prospect of a nude trip to one of Cornwall’s most popular tourist destinations,” he added.

As well as visiting Blue Reef, the naturists also went scuba diving and enjoyed a fishing trip during their week-long stay at Newperran Holiday Park.

Blue Reef Aquarium’s Steve Matchett said: “This will be the third time we have hosted a naturist event here at the aquarium.

“Our aquarists are incredibly enthusiastic about the aquarium and promoting awareness of the marine environment and I’m sure that any initial nerves will soon be forgotten and they’ll just get on with giving the guests a fantastic and highly entertaining visit,” he added.

Extra Color Enhancers in Koi Feeds, And Pellet Size

Excess color enhancers

Use of extreme color enhancers in meals could add to Yellowhead (which is fifty percent hereditary, with food potentiation) Male fish of the Shiro lesson (and others) could have a yellow, or “bone” tinted head. This is just even worse on fish that get color enhancers. However bear in mind, it’s the maleness of the fish and it’s genetics that set this in play, not the meals.

Pink skin – This is an urban legend. I have actually never ever viewed this even in fish supplied crazy amounts of shrimp oil experimentally. However, it still exists in the literature. If I ever before get a possibility to make you an electronic unretouched photo of a case of this I will, and I will allow you understand.

A quirk about pellet dimension

Small fish need small pellets that they could wholly engulf, yet they will spend time chasing after the biggest pellets. It would certainly be much better for the fish if they were given a tiny pellet they might completely swallow up. They could possibly fill their bellies rather than scraping off a dish over a prolonged time.

Can small fish eat huge pellet? Yes, yet that is only by nagging the huge pellet around the area of the pond as it softens in the water, and consuming off it like a huge peach.
